PRODUCTION AND PURIFICATION OF CELLULASE FROM CULTURE OF S.CEREVISIAE UNDER OPTIMUM FERMENTATION CONDITIONS ON ONION BROWN SKIN

Abstract

An increase in the global population is currently accompanied by increased agricultural productivity. This 
concurrently causes the generation of enormous amounts of agricultural waste that puts the sustainability of the 
ecosystem at risk. This study aimed to produce cellulolytic enzymes using local fungi isolates through the solid-state 
fermentation of some underutilized agro wastes: roasted groundnut skin, onion-brown skin. The major population of 
the isolates was identified as saccharomyces cerevisiae.  The maximum production of the cellulolytic enzymes 
(endoglucanase, exoglucanase, β-glucosidase) was observed at 96 h. The highest output of β-glucosidase and 
exoglucanase was at pH 4 while endoglucanase activity was highest at pH range 4 and 5. An increase in temperature 
increased the amount of cellulase production and the optimum temperature was observed at 40 °C. Activity of the 
cellulolytic enzymes follows Michaelis-menten hyperbolic curve using the three substrates. An increase in the 
inoculum size resulted in decreased activity of endoglucanase, exoglucanase on onion brown skin while β
glucosidase activity increased on the three substrate.
